SDS-PAGE Loading Buffer
SDS-PAGE Loading Buffer is the most commonly used sample buffer for SDS-PAGE. It is ideal for preparing protein samples to be separated in the SDS-PAGE electrophoresis system and allows for easy sample monitoring during electrophoresis.
SDS can bind to proteins to make a large amount of negative charge on the protein SDS complex. At this time, the charge of the protein itself is completely covered by SDS, eliminating the difference in charge of various proteins. SDS can also break intramolecular and intermolecular. The hydrogen bond, which destroys the secondary and tertiary structure of the protein molecule, beta-Mercaptoethanol can break the disulfide bond between the cysteine residues, destroy the protein structure, and eliminate the difference between the protein structures. In the final protein (subunit) with no charge and structural differences, the electrophoresis speed is only related to its molecular weight. Bromophenol blue is used as an indicator for electrophoresis and can roughly indicate the time at which electrophoresis ends.
The loading buffer contains SDS, Bromophenol Blue with beta-Mercaptoethanol in recommended concentrations. The buffer is supplied in 5x concentrate and should be diluted prior to use in electrophoresis.
