Principle
- PPLO broth base is composed of beef heart infusion form, peptone, and sodium chloride. Beef heart infusion form and peptone provide nitrogen, vitamins, amino acids, carbon, and other essential nutrients.
- Sodium chloride maintains the osmotic balance. The media must be fortified with a mycoplasma supplement composed of potassium tellurite, yeast extract, desiccated horse serum, and penicillin.
- Potassium tellurite inhibits many gram-negative bacteria. Yeast extract provides nitrogen for replication and growth.
- Horse serum is the source of cholesterol, a mycoplasma growth stimulant. Penicillin inhibits gram-positive as well as gram-negative bacteria.
