Principle
- PPLO broth base is composed of beef heart infusion form, peptone, sodium chloride, and crystal violet. Beef heart infusion form and peptone provide nitrogen, vitamins, amino acids, carbon, and other essential nutrients. Sodium Chloride maintains the osmotic balance.
- Crystal violet inhibits most gram-positive bacteria. The media must be fortified with a mycoplasma supplement composed of potassium tellurite, yeast extract, desiccated horse serum, and penicillin.
- Potassium tellurite inhibits many gram-negative bacteria. Yeast extract provides nitrogen for replication and growth. Horse serum is the source of cholesterol, a mycoplasma growth stimulant. Penicillin inhibits gram-positive as well as gram-negative bacteria.
