Principle
Phenol Red Broth Medium is formulated as per Vera (1950) and is composed of protease peptone, meat extract, mannitol, sodium chloride, and phenol red. The media is used to determine the fermentation reaction of carbohydrates for the differentiation of microorganisms. The differentiation of microorganisms is based on their ability to ferment the specific carbohydrate (Mannitol), with the production of acid or acid and gas. Protease peptone and meat extract serve as sources of carbon and nitrogen and other essential growth factors. Sodium chloride maintains osmotic equilibrium. Mannitol is fermentable sugar. Phenol red is the pH indicator, which turns yellow at acidic pH due to the fermentation of mannitol. Gas formation is confirmed by bubbles trapped in Durhams tubes.
