PCR Based Libraries are rapid & easier to construct but is only possible if screening of the DNA fragments is performed by nucleic acid hybridization & is not applicable when functional analysis of the encoded protein is required. PCR based libraries are constructed using combination of revised transcriptase & polymerase chain reaction.
