Principle
- Mitis salivarius agar is composed of tryptose, protease peptone, dextrose, sucrose, dipotassium phosphate, tyrpan blue, crystal violet, and agar. Tryptose and protease peptone are sources of carbon, nitrogen, vitamins, and minerals.
- Dextrose and Sucrose are carbohydrate sources. Dipotassium phosphate provides phosphate. Trypan blue is an acidic, blue diazo dye, gives a blue color to colonies
- Crystal violet is a basic dye that inhibits many gram-positive organisms. Agar is the solidifying agent. Potassium Tellurite (not provided with medium) inhibits most gram-negative bacilli and most gram-positive bacteria except streptococci.
