Hoechst 33342/PI Dual Staining Assay - Cell Detection Reagent
Hoechst 33342/PI Dual Staining is based upon fluorescent detection for the compacted state of the chromatin in apoptotic cells.
The kit employs Hoechst 33342 and Propidium Iodide dyes; assaying normal, apoptotic, and dead cells.
Hoechst 33342 is a cell permeable fluorescent minor groove-binding probe for DNA, and through the living cell membrane. It stains the condensed chromatin in apoptotic cells more brightly than the chromatin in normal cells. Propidium iodide (PI) has been widely used as a fluorescent stain for DNA in cells; it specificity binds double-stranded nucleic acids in apoptosis, necrosis and fixed cells, but cannot enter the normal living cell. It is only permeant to dead cells. The staining pattern resulted from the simultaneous use of these dyes makes it possible to distinguish normal, apoptotic, and dead cell populations by flow cytometer and fluorescence microscopy. The population should be separated into three groups: live cells will show only a low level of fluorescence; apoptotic cells will show a higher level of blue fluorescence, and dead cells will show low-blue and high-red fluorescence.
