Different formats like direct, indirect or sandwich can be developed. Direct & indirect ELISA both immobilize the antigen on assay plate & then use either a labeled primary antibody (direct) or primary antibody & labeled secondary antibody (indirect) to detect the antigen. Sandwich ELISA is considered most robust format because the antigen is 'sandwiched' between two primary antibodies & then detected using labeled secondary antibody.
