Diff-Quik staining Kit
Diff-Quick staining is mainly used for blood cell smear, bone marrow smear, vaginal secretion smear, and exfoliated cell smear. Diff-Quick staining is ideal for batch dip dyeing with clear background and no sediment.

Product Description
Diff-Quik staining is a rapid staining method based on Wright staining. It is one of the common staining
methods in cytology. The staining solution is prepared by the rapid staining method recommended by
the World Health Organization (WHO). It is similar to Wright Stain and is improved by using the
principle of Romanowsky Stain technology. The staining result is also very similar to the Wright staining
solution. The required time is extremely short, and the dyeing can be completed within 90s.
Diff Rapid Staining Solution contains fixative, mainly used for blood cell smear, bone marrow smear,
vaginal secretion smear, exfoliated cell smear. Diff-Quik staining is ideal for batch dip dyeing with clear
background and no sediment.
Protocol
1. Prepare blood smears or bone marrow smears by conventional methods, dry them naturally or
treat them with an alcohol lamp flame, and then use Diff-Quik Fixative to fix them. Set for 20s.
2. Diff-Quik I staining for 5-10s (lift the slide up and down 2-3 times to make the dyeing solution
evenly distributed), take it out immediately.
3. Diff-Quik II staining for 10-20s (lift the slide up and down 2-3 times to make the dyeing solution
evenly distributed), take it out immediately.
4. Immediately after washing with water, observe under the microscope while wet.
5. Bring the films deemed valuable after observation to the laboratory, transparent with xylene,
seal and store.
Result:
Nucleus, White Blood Cells- Dark Blue
Stroma, lymphocytes- Purple
Cytoplasm, Red Blood Cells- Pink
Important Notes:
1. Diff-Quik Fixative is a special fixative for Diff-Quik. If it is used in large quantities, it can be
replaced by methanol.
2. The blood smear or bone marrow smear should be evenly thick so as not to affect the staining
effect.
3. Blood cell smear staining requires fresh whole blood or EDTA anticoagulant blood.
4. Bone marrow smear staining requires the prepared smear to be quickly shaken or fan-dried in
the air to prevent the cells from shrinking and deforming or due to humid air for hemolysis,
high temperature or fire baking should not be used to dry the smear.
5. Vaginal secretion smear staining requires that fresh specimens be smeared with flame or
alcohol as soon as possible to prevent cell deformation.
6. Exfoliated cell smears can be fixed by natural drying or fixative solution.
7. Do not remove the dye solution or wash the smear directly during staining of the smear. Don't
pour out the dye solution first, so as not to deposit the dye on the smear.
8. The staining solution can be reused, but not repeated multiple times. If there is sediment, it
should be filtered and used.
9. If the dyeing is too deep, it can be decolorized with methanol or alcohol, preferably without
counterstaining.
10. If the dyeing is too dark or too light, adjust the dyeing time or working solution concentration.
11. The pH value has a certain influence on the staining; the glass slide should be clean and free of
acid and alkali pollution, so as not to affect the staining effect.
