Aprotinin is widely used in protein extraction processes and plays an important role in inhibiting protease-decomposing proteins. Aprotinin inhibits kallikrein in trypsin, plasmin, plasma, and tissue. Serine-active proteases such as plasmin and plasma kallikrein play key roles. Aprotinin inhibits the active portion of the serine on the enzyme, but binds to a different protease, and the inhibitory effect is slightly different. The binding to trypsin is the strongest, and the binding to human plasmin is very low, and the complex with human plasma kallikrein is rather weak. The antifibrinolytic effect of aprotinin not only protects the direct substrate (fibrin) from degradation by fibrinolytic enzymes, but also protects fibrinogen in plasma and alpha 2-globulin in serum.
